Om Micro-Propagation Techniques of Kinnow mandarin (Citrus deliciosa)
Research work on Micro-propagation techniques of Kinnow mandarin was carried out at the TCL, SKRAU, Bikaner. Several aspects such as establishment of axenic culture, axillary bud proliferation, rooting in micro-shoots and in-vitro shoot tip grafting formed various parts of the entire investigation. The mercuric chloride (0.1 %) is the best treatment for establishment of aseptic culture. Among auxin used singly for shoot regeneration parameters lower levels of NAA were found better than IAA. Among cytokinin, BAP was found better for shoot multiplication whereas, kinetin was better for length of shoots. The higher level of cytokinins had negative effect on all parameters. The combination of cytokinins with auxin had better synergetic effect. Lower levels NAA and IBA were found better as compared to higher levels. The synergetic effect of both had pronounced effect. In in-vitro shoot tip grafting experiments, kinnow/Carrizo (12 days old) seedling was found better rootstock than others. Among PGRs 2,4-D was better as pre treatment for increasing success of STG. In green house, maximum survival of plantlets was recorded in Rough lemon and STG on carrizo.
